The AKTA pure 25L is a modular chromatography purification system developed by Cytiva (formerly GE Healthcare), specifically designed for rapid purification of biomolecules including proteins, peptides, and nucleic acids. This system is suitable for separation and purification of target products ranging from microgram to gram scale (maximum processing capacity up to tens of grams). Based on liquid chromatography separation principles, it utilizes differences in transport velocity between compounds in stationary and mobile phases to achieve separation, supporting various purification techniques including gel filtration chromatography, ion exchange chromatography, hydrophobic chromatography, and affinity chromatography.

Core Technical Features

The AKTA pure 25L adopts a modular design, comprising three key modules:

1. Liquid Delivery System:

   • Dual-piston pump system with flow rate range 0.25-25 mL/min (up to 50 mL/min in column packing mode)
   • Maximum pressure resistance 20 MPa, suitable for various chromatography columns
   • Optional sample pump (0.01-50 mL/min) for automatic sample injection

2. Detection System:

   • Multi-wavelength UV detector (U9-M), simultaneously monitoring three wavelengths from 190-700nm
   • Optional single-wavelength detector (U9-L, fixed at 280nm)
   • Conductivity detector (0-999 mS/cm) and pH monitoring module

3. Fraction Collection System:

   • Flexible F9-C fraction collector compatible with 24/48/96-well deep well plates and various collection tubes
   • Automatic collection by time, volume or peak recognition
   • Maximum collection capacity of 576 fractions
